高效液相色谱法测定人血浆中顺铂的含量

摘　　要：目的建立以高效液相色谱法测定人血浆中顺铂含量的方法。方法色谱柱为Hypersil-ODS2,流动相为0.25mol/L氯化钠溶液-甲醇(29∶71),流速为1.1ml/min,检测波长为254nm,柱温为38℃。结果顺铂的线性范围为0.2~10μg/ml(r=0.9998),平均回收率为100.72%(n=5),最低检测浓度为0.1μg/ml。结论本方法是检测顺铂血药浓度的一种有效方法。OBJECTIVE:To establish a HPLC method for the determination of cisplatin in human plasma.METHODS:The separation was performed on a Hypersil-ODS 2 column with column temperature at38℃.The mobile phase consisted of0.25mol/L sodium chloride-methanol(29∶71)with a flow rate of1.1ml/min and detective wavelength of254nm.RESULTS:The linear range of cisplatin was0.2～10μg/ml(r=0.9998).The average recovery of cisplatin was100.72%(n=5).The minimum concentration was detected at0.1μg/ml.CONCLUSION:The method is effective ...
