液相色谱-串联质谱法测定猪肉中的莱克多巴胺残留

摘　　要：建立了猪肌肉组织中莱克多巴胺的高效液相色谱-串联质谱检测方法.用0.1 mol/L高氯酸提取猪肌肉组织中的药物,经阳离子固相柱净化,采用正电喷雾离子源,在多反应监测(MRM)模式下进行分析,三对定性离子对分别为:m/z 301.90>136.00、301.90>164.10 和 301.90>284.00,定量离子对为301.90>284.00.在0.1～20 ng/mL浓度范围内,呈良好的线性关系,相关系数r＞0.99.在0.5、2、10 μg/kg三个添加水平,平均回收率为75.2%～97.6%,日内变异系数在5.5%～12.3%,日间变异系数在7.8%～14.7%,检测限为0.1 μg/kg.A method for determination of ractopamine in swine muscle with liquid chromatography—tandem mass spectrometry was established.The drug in swine muscle was extracted with 0.1mol/L perchloric acid,purified by MCX SPE columns and detected by positive electrospray ionization and multiple reaction monitoring(MRM) of three parent-daughter ion pairs: m/z 301.90>136.00、301.90>164.10 and 301.90>284.00.The ion pair of m/z 301.90>284.00 was used for quantitation.There was a good linear correlation(the coefficient of correlation was above 0.99) between the peak areas and concentration of ractopamine with the range from 0.1 to 20 ng/mL.Recoveries ranged from 75.2% to 97.6% with coefficients of variation from 5.5% to 14.7%.The limit of detection is 0.1 μg/kg.
