摘 要:目的寻找尘埃暴露人员血清生物标志物。方法利用表面加强激光解析电离飞行时间质谱(Surface enhanced laser desorption ionization-time of flight-mass spectrometry,SELDI-TOF-MS)技术检测60例尘埃暴露人员的血清蛋白质指纹图谱(接触尘埃未患病者为0组20例,可疑尘埃者为0+组20例,患Ⅰ期硅沉着病者为Ⅰ期组20例)。采用美国Cipher-gen生物公司研制的新一代弱阳离子交换芯片(CM10)和蛋白质芯片阅读仪读取数据,获得的蛋白质谱采用Ciphergen公司的Ciphergen protein chip软件和Biomarker wizard软件分析。结果尘埃暴露人员各组间对比,发现M/Z为2938、3269、3821、4095、5071、54835、640的7个标志分子在尘埃暴露人员血清中差异表达,相对含量变化具有统计学意义(P<0.05)。其中M/Z为5071、5483和5640的分子在尘埃暴露人员0组和0+组中均为低表达,而在Ⅰ期组中高表达。结论M/Z为5071、5483和5640的分子可能是尘埃暴露人员血清生物标志物。Objective To study the specific biomarkers in serum of silica-exposed workers.Methods Surface enhanced laser desorption ionization-time of flight-mass spectrometry(SELDI-TOF-MS) was used to search the specific biomarkers in serum samples from 60 silica-exposed workers,including 20 with no silicosis(0 group),20 with suspected silicosis (0+ group),and 20 with stage I silicosis(stage I group).Serum samples were analyzed using the new generation weak cationic exchange chip(CM10,Ciphergen Biosystems,USA),and the protein fingerprints automatically collected using Ciphergen ProteinChip Biology System(PBSII-C).The data were analyzed using Ciphergen ProteinChip and BioMarker Wizard software.Results There were expressions of 7 proteins,M/Z 2 938,3 269,3 821,4 095,5 071,5 483,and 5 640,among the silica-exposed workers(P<0.05).Of them,M/Z 5 071,5 483 and 5 640 were lowly expressed.in 0 and 0+ groups but highly expressed in I stage group.Conclusions M/Z 5 071,5 483,and 5 640 may be the biomarkers for the silica-exposed workers.
