肾康注射液对系膜细胞肥大及p21^(CIP1/WAF1)、p27^(KIP1)表达的影响

摘　　要：目的观察肾康注射液对高糖培养大鼠肾小球系膜细胞肥大及p21^(CIP1/WAF1)(p21)、p27^(KIP1)(p27) mRNA及蛋白质表达的影响。方法将体外培养的大鼠肾小球系膜细胞分为6组,分别为正常组、甘露醇组、高糖组、高糖加肾康注射液高、中、低剂量组,用[~3H]胸腺嘧啶核苷([~3H]TdR)及[~3H]亮氨酸([~3H] Leu)掺入法反应细胞DNA及蛋白质的合成情况,RT-PCR、Western blot检测p21mRNA及p21、p27蛋白质表达情况。结果高糖培养肾小球系膜细胞蛋白质合成增加,DNA合成减少,p21mRNA及蛋白质、p27蛋白质表达增加；肾康注射液组与高糖组比较,蛋白质合成减少,DNA合成增加,p21mRNA及蛋白质、p27蛋白质表达降低。结论肾康注射液可能部分通过降低p21、p27过表达来阻止高糖诱导肾小球系膜细胞的肥大反应。Objective To investigate the effect of Shenkang Injection(SI)on hypertrophy and expressions of p21 and p27 in glomerular mesangial cells of rats cultured in high glucose fluid.Methods Cultured rat glomerular mesangial cells were divided into 6 groups: the normal control group,mannitol-treated group,high glucose-treated group,high,middle and low dose SI treated groups.The synthesis of cell protein and DNA were determined with in- corporation of^3 H-TdR and^3 H-Leu.The changes of p21 mRNA and expressions of p...
