摘  要:目的表皮生长因子(EGF)对先天性膈疝(CDH)大鼠肺发育不良具有明显的改善作用,本研究探讨EGF对大鼠CDH模型肺发育中相关活性物质的影响,进而了解EGF可能的作用机制。方法建立大鼠CDH模型,将实验大鼠随机分为正常组、CDH组、CDH+EGF组,采用免疫组织化学方法检测各组胎肺组织中表皮生长因子受体(EGFR)、甲状腺转录因子-1(TTF-1)以及表面活性蛋白B(SP-B)等活性物质的表达情况。结果EGFR,TTF-1,SP-B在各实验组中均有表达。与正常组相比,EGFR在CDH组和CDH+EGF组中表达强度均有显著性差异(P<0.05)。SP-B在CDH组与正常组,CDH+EGF组与CDH组中表达强度的比较均有显著性差异(P<0.05)。TTF-1在正常组、CDH组和CDH+EGF组的两两比较也均有显著性差异(P<0.05)。在不同EGF剂量组的胎肺组织中EGFR、SP-B和TTF-1均有表达,各指标在不同EGF剂量组间的表达强度均有显著性差异(P<0.05),且EGFR、SP-B和TTF-1的表达随EGF作用剂量的增加呈现逐渐增加的趋势(P<0.05)。结论表皮生长因子对CDH胎肺组织EGFR,TTF-1,SP-B的表达部位有影响,并且对其表达强度有明显上调作用。Objective To explore the effects of epidermal growth factor(EGF)on surfactant-associated protein in pulmonary development in congenital diaphragmatic hernia(CDH)model in tats,in order to further understand EGFaction in pathogenetic mechanism.Methods CDH model of rats was constituted by Notrofen intragastric perfusion,22 SD female rats was rondomly divided into three groups;control group,CDH group and CDH+EGF group before the experiment animals model establishment..Epidermal growth factor receptor(EGFR)antibody,thyroid transcription factor-1(TTF-1)and surfactant protein-BC(SP-B)antibody were determined by immunohistochemical staining respectively in each group..in order to clarify change for expressions of the three factors.Results The expression of EGFR,TTF-1 and SP-B could be detected in each subgroup of EGF+CDH.In contrast with control group,existed significant differences for the EGFR expression in CDH group and EGF+CDH group(P<0.05).Compared with CDH group,SP-B expression was statistically significant for EGF+CDH and control groups(P < 0.05).The expression of TTF-1 was statistically significant between any two groups.At the same time,the expressions of EGFR,SP-B and TTF-1 up-regulation with increasing EGF dosages(P < 0.05).Conclusions EGF could affect the expression locations and extensities of EGFR,SP-B and TTF-1 in the CDH lung.
