摘 要:目的研究口服生长抑素(SS)基因疫苗对小肠局部细胞免疫的影响。方法应用SS基因疫苗通过两种方式(试验组Ⅰ和试验组Ⅱ)灌胃小鼠,首次免疫后第3、5和7周分别扑杀小鼠,应用HE染色、亲和免疫组化和甲苯胺蓝染色的方法分别显示小肠上皮内淋巴细胞(iIEL)、固有层CD4+T细胞和肥大细胞的分布及数量变化。结果两个试验组iIEL数量比对照组有极显著增加(P<0.01);首次免疫后小肠固有层CD4+阳性细胞数量增加,但不显著;首次免疫后第3周空肠中肥大细胞数量与对照组相比有极显著性增加(P<0.01)。结论本试验结果提示了SS基因疫苗能提高小鼠肠道细胞免疫功能。Objective To investigate the effects of Somatostatin(SS) DNA vaccine on local immunological function of small intestine.Methods Mice were immunized with SS DNA vaccine orally by two feeding regimes(treatmentⅠ and treatmentⅡ),and then were sacrificed at week 3,5,and 7.The distribution and the change of iIEL cells,lamina propria CD4+ T cells,and mast cells in mice duodenum and jejunum were studied with Hemetoxylin and Eosin Staining (HE),immunohistochemical methods,and Toluidine blue staining(TBS).Results The number of iIEL(treatmentⅠ and treatmentⅡ) in duodenum and jejunum was increased significantly(P < 0.01) after oral immunization compared with control group.The lamina propria CD4+ T cells and the mast cells in jejunum were also increased(P > 0.05 and P < 0.01,respectively) at 3 weeks after oral immunization compared with control group.Conclusion Results suggest that oral SS DNA vaccine can improve the mucous immune function in mice intestine.
