摘  要:目的 观察大鼠前列腺痛模型中L5~S2脊髓背角胶质原纤维酸性蛋白(GFAP)、肿瘤坏死因子-α(TNF-α)和诱导型NO合成酶(iNOS)蛋白表达的变化.方法 取30只SPF雄性大鼠完全随机分为实验组和对照组(设0、6、12d 3个时相,分为5组,每组6只),通过前列腺完全弗氏佐剂(CFA)和生理盐水注射制作大鼠前列腺痛模型和对照组,分别提取L5~S2脊髓背角组织蛋白,Western印迹检测脊髓背角GFAP、TNF-α、iNOS的表达.结果 慢性前列腺痛大鼠中L5~S2脊髓背角中GFAP、TNF-α、iNOS表达在6d和12d组均明显高于各自正常对照组和0d组,差异有统计学意义(P<0.01),GFAP 12d组高于6d组,差异有统计学意义(P<0.05),iNOS的表达高峰在6d组,差异有统计学意义(P<0.01).结论 慢性前列腺痛可以引起L5~S2脊髓星形胶质细胞活化和TNF-α分泌增多及iNOS活性增强,表明慢性前列腺痛可以导致L5~S2脊髓中枢继发性的炎性改变,可能与前列腺痛的持续和泛化有密切关系.Objective To investigate the express of GFAP,TNF-α and iNOS in posterior horn of L_5~S_2 spinal cord of rat chronic prostate pain model.Methods The rat chronic prostate pain model was made by injecting CFA to prostate.The alteration of the express of GFAP,TNF-α and iNOS in the posterior horn of L_5~S_2 spinal cord were observed with Western-blot.Results The express of GFAP,TNF-α and iNOS in posterior horn of the L_5~S_2 spinal cord of the rat prostate pain model was obviously higher than that of the control at 6d,12d and 6d,12d were higher than 0d(P<0.01),moreover,the express of GFAP at 6d was lower than at 12d(P<0.05) and the peak value of iNOS express was at 6d(P<0.01) in experiment groups.Conclusion The chronic prostate pain can result in the activation of astrocytes and more express of TNF-α and iNOS in L_5~S_2 spinal cord,so chronic prostate pain may lead to the the secondary inflammation of L_5~S_2 spinal cord,which is probably correlated with the maintenance and enlargement of prostate pain.
