摘 要:目的通过对纳米羟基磷灰石(nanosize-hydroxyapatite,nHA)的毒性检测,初步探讨生物材料分子生物相容性研究的必要性。方法通过形态学观察、四甲基偶氮唑蓝(methl thiazolyl tetrazolium,MTT)实验及彗星电泳检测nHA的毒性。结果倒置显微镜观察显示不同浓度的nHA浸提液对体外培养细胞的形态无明显影响;MTT实验表明nHA对细胞生长和增殖无明显抑制作用,不同浓度浸提液的细胞毒性均为1级,无毒,与生长良好的L-929细胞形态观察结果相符合;彗星电泳结果显示随nHA浸提液浓度和时间的递增彗星率逐渐增大。结论nHA具有良好的细胞相容性,但对L-929细胞DNA可能有损伤作用,提示可能有遗传毒性作用,因此检测纳米材料分子生物相容性非常必要。Objective To detect the toxicity of nanosizehydroxyapatite(nHA) and study the responsibility on biocompatibility of nano-materials.Methods The morphological change was observed by invert microscopy.MTT assay was used to evaluate the cytotoxicity in vitro.The effects on DNA damage of cultured L-929 cell were observed through comet electrophoresis.Results MTT assay showed that various concentrations leaching liquors of nHA had neither reliable effect on cell morphology,nor significant inhibition on cell growth and proliferation.The cytotoxicity gradation of various concentrations of the extract of nHA was 1 degree,the same results were discovered on the morphological change by invert microscopy,there was no cytotoxicity to leaching liquors of nHA.Comet electrophoresis detecting showed that comet ratio of L-929 cells induced by leaching liquors of nHA went up gradually with the increase of dosage and time.Conclusion nHA has good cytocompatibility,but maybe have damage-contribution to L-929 cells in DNA level,implying remarkable genotoxicity,therefore,it is vital to evaluate molecular biocompatibility on nano-materials.
