摘 要:目的探讨低强度脉冲半导体激光对大鼠成骨细胞增生和分化的影响。方法从新生SD大鼠颅盖骨分离成骨细胞,体外培养。用低强度脉冲半导体激光(650nm,2mW)照射细胞5min(1.14J/cm2,A组)和10min(2.27J/cm2,B组),并与未被照射的细胞(C组)比较。采用MTT法和ELISA法检测细胞数量和碱性磷酸酶(ALP)含量,作为成骨细胞增生和分化的指标。结果经低强度脉冲半导体激光照射后第7天,A组和B组的细胞增生数量分别较C组平均增加7.7%和7.6%(n=8,P<0.0001);照射后第21天A组和B组的ALP含量(U/L)分别较C组平均增加81.7%和66.4%(n=8,P<0.0001);细胞增生数与ALP含量A组和B组组间差异无显著意义(P>0.05)。结论低强度脉冲半导体激光能明显促进体外培养的大鼠成骨细胞的生长、分化和成熟,对骨重建有调节作用,对防治骨质疏松症等骨病可能是一种有效而安全的新疗法,值得深入研究。Objective To investigate the effect of low-intensity pulsed diode laser irradiation on proliferation and differentiation of rat osteoblasts in vitro. Methods Osteoblasts were isolated from calvariae of newborn SD rats, and suspended in culture medium. After stimulation by a low-intensity pulsed diode laser irradiation (650nm,2mW) for 5min (1.14J/cm2, Group A) and 10 min(2.27J/cm2,Group B), the cellular proliferation and alkaline phosphatase (ALP) level of osteoblasts in above mentioned 2 groups and the Group C (non-lased cultures as the control) were examined by MTT and ELISA assay respectively. Results The mean value of osteoblast amount in the Groups A and B was 7.7% and 7.6%,more than that of Group C (n=8, P<0.0001), and the mean value of ALP (U/L) account in the Groups A and B was 81.7% and 66.4%,more than that of the Group C (n=8,P<0.0001) respectively, but there was no significant difference statistically between the Groups A and B (P>0.05). Conclusions Low-intensity pulsed diode laser irradiation can significantly promote the proliferation, differentiation and maturation of rat osteoblasts in vitro, and it may be a potential method in treatment and prevention of bone diseases such as osteoporosis by its beneficial effects on bone remodeling.
