防己茯苓汤加减对大鼠肾系膜细胞增殖及基质金属蛋白酶-2的影响

摘　　要：目的:观察防己茯苓汤加减方是否具有干预肿瘤坏死因子诱导后肾小球系膜细胞增殖的作用及对基质金属蛋白酶MMP-2的影响。方法:采用大鼠肾小球系膜细胞培养进行体外研究,用肿瘤坏死因子(TNF-α)刺激系膜细胞增殖,再加入不同浓度防己茯苓汤加减方提取液对细胞进行干预,用MTT法测定细胞增殖,流式细胞仪分析细胞周期,用ELISA法测定细胞培养液中肾小球系膜细胞基质金属蛋白酶(MMP-2)的含量。结果:加入防己茯苓汤加减方提取液后,系膜细胞的增殖程度受到显著抑制(P<0.05),流式细胞仪DNA图示S期细胞数显著减少(P<0.05)。细胞培养液中MMP-2的表达增强(P<0.05)。结论:防己茯苓汤加减方对肿瘤坏死因子诱导后肾小球系膜细胞的增殖具有抑制作用,并可影响肾小球系膜细胞MMP-2的表达,进而调整细胞外基质(ECM)的合成与降解。Objective:To investigate the effects of Fangji Fuling Tang on preventing glomerular mesangial cells' proliferation and affecting the expression of MMP2.So as to provide experimental evidence for using Fangji Fuling Tang to cure Chronic kidney disease.Method:Glomerular mesangial cells were cultured.TNF-α was used as a stimulation to make the cells to proliferate.Then Fangji Fuling Tang at different doses was incubated with the cells.Cell vitality was evaluated and qualified by MTT.Cell cycle were analysed by Flow CytoMeter(FCM).Secretion of MMP-2 expression was measured by ELISA.Results:Gtomerular mesangial cells' proliferation was inhibited (P<0.05),the DNA diagram of Flow CytoMeter showed that the cell quantity in stage 'S' was reduced(P<0.05),the expression of MMP-2 was markedly increased(P<0.05)in Fangji Fuling Tang groups.Conclusion:Fangji Fuling Tang may inhibit glomerular mesangial cells' proliferation and influence the synthesis and degradation of extracellular matrix.
