高效液相色谱法比较不同工艺制备的黄连解毒汤中盐酸小檗碱含量

摘　　要：目的建立反相高效液相色谱法测定并比较传统饮片与现代免煎饮片制备的黄连解毒汤中盐酸小檗碱的含量。方法色谱柱Hypersil ODS(4.6×250mm,5μm);保护柱:Zorbax ODS(4.6×12.5mm,5μm);流动相:MeOH-H_2O-三乙胺(50:50:4);流速:0.8mL/min,UV检测波长346nm。结果盐酸小檗碱在6.25~200μg·mL^(-1)间有良好的线性关系,r=0.9996,日内、日间RSD分别为0.7%、0.9%,传统饮片与免煎饮片配制的汤剂盐酸小檗碱回收率分别为92.0%、90.1%。结论该方法简便可行,结果可靠。OBJECTIVE To establish HPLC method for determining the content of berberine HCl in Huanglian Jiedu Decoction and comparison with the traditional water and exempt decoction process.METHOD The column was Hypersil-ODS.The mobile phase was methanol-water-triethylamine(50:50:4),detecting wavelength was 346 nm.RESULTS The method is linear in the range of 6.25~200.00μg·mL^(-1),r = 0.9996,The intra-day and inter-day RSD were 0.7%、0.9%.The recovering rate of traditional water and exempt decoction process were 92.0%、90.1%.CONCLUSION The method is simple and practical,The results are precise.
