摘 要:目的 观察中药免疫增强剂和化学免疫抑制剂对吡喹酮杀血吸虫效果的影响。 方法 日本血吸虫感染昆明小鼠后随机分成四组 ,对照组、吡喹酮治疗组、氢化可的松 (免疫抑制剂 ) +吡喹酮治疗组、中药 (免疫增强剂 ) +吡喹酮治疗组 ,分别采取相应不同的治疗方式 ,于吡喹酮治疗后 2周剖杀小鼠 ,观察减虫及减卵效果 ;并于吡喹酮治疗前采血 ,用ELISA方法测定特异性抗血吸虫成虫抗原 (AWA)抗体水平。 结果 与对照组相比 ,吡喹酮治疗组、氢化可的松 +吡喹酮治疗组、中药 +吡喹酮治疗组减虫率分别为 5 3 .2 2 % ,3 5 .61% ,69.16% ;每克肝卵减少率分别为 47.0 8% ,3 1.2 7% ,63 .2 8%。氢化可的松 +吡喹酮治疗组、中药 +吡喹酮治疗组与吡喹酮治疗组相比 ,减虫与减卵效果统计学有显著差异 (P<0 .0 5 )。且各组特异性抗AWA抗体水平高低与减虫、减卵效果相一致。 结论 进一步证明吡喹酮杀血吸虫作用具有免疫依赖性 ,而且证实中药免疫增强剂具有增强宿主特异性免疫应答 ,提高吡喹酮杀虫效果的作用。Objective To observe the influence of immunoenhancer and immunosupression on praziquantel killing Schistosoma japonicum. Methods Mice infected with Schistosoma japonicum were randomly divided into four groups: control group,praziquantel treat group,hydrocortisone (immunosupression) plus praziquantel treat group and Chinese medicine herb ( immunoenhancer ) plus praziquantel treat group. The mice of different groups were given corresponding treament and were dissected after two weeks of praziquantel treatment. The worm burden and liver eggs per gram ( LEPG ) were counted. Obstaining blood before Praziquantel treatment,the level of specific anti-AWA antibody were detected by ELISA. Result The reduction in worm burden in praziquantel treat group,hydrocotisone plus praziquantel treat group and Chinese medicine herb plus praziquantel treat group was 53.22%,35.16% and 69.16% respectively,and LEPG was 47.08%,31.27% and 63.28% respectively. Compared with praziquantel treat group,the reduction in worm and egg were significantly different in Hydrocortisone plus raziquantel treat group and Chinese medicine herb plus praziquantel treat group. The level of specific anti-AWA antibody responded with reduction in worm and egg. Conclusion The results had not only further revealed that praziquantel against Schistosoma japonicum was dependent on immunity but also proved that Chinese medicine herb immunoenhancer selected by us could enhance host specific response and improve praziquantel killing of Schistosoma japonicum.
