摘 要:目的 :探讨大黄苷元及其单体对大鼠脑缺血损伤的保护作用。方法 :线栓阻塞大鼠大脑中动脉 ,制备局灶性脑缺血模型 ,观察大鼠神经症状 ,测定脑梗塞面积 ,检测脑组织TNF -α变化。结果 :模型组大鼠较假手术组的神经症状评分增高 ,脑梗塞面积增大 ,脑组织TNF -α水平增高 ;与模型组比较 ,大黄苷元、大黄素、芦荟大黄素各剂量组神经症状评分降低 ,川芎嗪组、大黄苷元和大黄素高剂量组、芦荟大黄素低剂量组、大黄酸高、中剂量组和大黄酚高、中剂量组脑组织梗塞面积减小 ,大黄苷元高、中剂量组脑组织TNF -α降低 ;与川芎嗪组比较 ,大黄素高剂量组神经症状评分降低 ,大黄苷元和大黄素高剂量组、芦荟大黄素低剂量组、大黄酸高、中剂量组和大黄酚高、中剂量组脑梗塞面积减小 ,大黄苷元高、中剂量组脑组织TNF -α降低 ;大黄素高剂量组神经症状评分均较大黄苷元高剂量组降低 ,大黄苷元高剂量组较芦荟大黄素和大黄素甲醚高剂量脑梗塞面积减小 ,脑组织TNF -α降低 ;大黄素高、中剂量组较其低剂量组评分降低。结论 :大黄苷元及其单体对脑缺血损伤具有保护作用 ,其作用机制可能与其降低脑缺血大鼠的TNFObjective:To study the protective effects of Rhubarb Aglycone and its monomers on rats with cerebral ischemia.Method: After the local cerebral ischemic models of cerebral middle artery occlusion were built up with nylon thread, the observation was made on changes of the rats neural symptoms, and the area of brain occlusion and the levels of brain TNF-α were measured. Results: Compared with the sham-operated group, the scores of neural symptoms and the area of brain occlusion in the model group were higher, and the level of brain TNF-α increased. Compared with the model group,the scores of neural symptoms were lower in each of the three groups of Rhubarb Aglycode, Emodin and Physcion; the area of brain occlusion shrinked in the group of Lgustrazin, Rhubarb, Aglycone (high dosage), Emodin (high dosage), Physcio(low dosage), Aloe-emodin (high and medium dosage) and Rhein (high and medium dosage); the levels of brain TNF-α in the group of Rhubarb Aglycone (high and medium dosage) decreased. Compared with the Lgustrazin group, the scores of neural symptoms in the group of Emodin (high dosage) decreased , and the area of brain occlusion shrinked in the group of Rhubarb Aglycone (high dosage), Emodin (high dosage), Physcion (low dosage), Aloe-emodin (high and medium dosage), Rhein (high and medium desage); the levels of brain TNF-α in the group of Rhubarb Aglycode (high and medium dosage) decreased. The evaluating score of neural symptoms in the group of Emodin (high dosage) was lower than that of the Rhubarb Aglycone (high dosage); the area of brain occlusion in the group of Rhubarb Aglycone (high dosage) reduced compared with that of the Emodin (high dosage) and Physcion (high dosage), and the level of brain TNF-α decresed. The evaluating score of neural symptoms in the group of Emodin (high and medium dosage) were lower than that of the low dosage.
