摘　　要：目的体外研究高糖刺激人肾脏间质成纤维细胞时肝细胞生长因子(HGF)表达,同时进一步观测了黄芪延缓糖尿病肾病进展是否与影响 HGF 表达相关。方法将人肾脏间质成纤维细胞培养于不同浓度葡萄糖中,分不同时间段采用 RT-PCR 方法检测 HGF mRNA 转录水平,ELISA 方法检测 HGF 蛋白分泌。之后用黄芪含药血清刺激细胞,观察 HGF 表达情况。结果高浓度葡萄糖培养的细胞早期就有 HGF 表达,之后表达逐渐下降。说明 HGF 表达与高糖作用存在相关性。结果进一步证实,黄芪含药血清可以显著刺激细胞表达 HGF。结论黄芪可以通过诱导 HGF 的产生,起到抗纤维化作用,从而延缓糖尿病肾病的进展。Objective To study in vitro the hepatocyte growth factor(HGF)expression in human renal mesenchymal fibroblast under high-glucose stimulation,and to explore whether the effect of Radix Astragali(RA)in postphoning the progression of diabetic nephropathy is related with its action on HGF.Methods Human renal mesenchymal fibroblast was cultured in media with different concentration of glucose.HGF mRNA transcription levels in various time points were determined by RT-PCR,and HGF protein secretion was detected by ...
