摘 要:采用RT PCR扩增的方法 ,从新疆野生植物犁苞滨藜中克隆获得 1.7kb的cDNA片段 ,测序和序列分析表明该基因片段包含NHX基因完整的读码框架。序列同源性分析结果显示犁苞滨藜NHX基因与滨藜NHX基因同源性高达 95 % ,与碱蓬同源性达到 86 % ,与柑桔同源性为 88% ,与拟南芥同源性为 84 % ,表明它是植物中高度保守的一种基因 。The NHX gene encodes functional protein that may play an important role in plant salt tolerance. To clone the NHX from the wild plant Atriplex dimorphostegia in Xinjiang into a T-vector, designed primers were used to amplify 1.7 kb NHX cDNA fragment by RT-PCR. Sequence analysis revealed that the cloned NHX fragment of Atriplex dimorphostegia contained entire NHX coding region with 95%,86%,88% and 84% identity comparing with Atriplex gemelini, Suaeda maritima, Citrus×paradisi and Arabidopsis thaliana, respectively. This analysis suggested that NHX gene was highly conserved in terms of evolution in plant; and it also suggested that the NHX gene of Atriplex dimorphostegia was similar to that of Arabidopsis. It may be of great importance in improvement of the plant salt tolerance and breeding of crop.
