健骨二仙丸含药血清对成骨细胞分化及Ⅰ型前胶原mRNA表达的影响

摘　　要：目的:在临床研究和动物实验的基础上,进一步探讨健骨二仙丸对成骨细胞分化的影响。方法:分离、培养人的原代成骨细胞,采用 PNPP 法、RIA 法、茜素红染色法以及定量 RT-PCR 之法分别检测碱性磷酸酶活性、骨钙素含量、矿化结节形成和Ⅰ型前胶原 mRNA 的表达,从而了解健骨二仙丸对成骨细胞分化的影响。结果:健骨二仙丸中、高剂量能显著增加碱性磷酸酶的活性及矿化结节形成的数量,促进成骨细胞骨钙素和Ⅰ型前胶原 mRNA 的表达(P<0.01,P<0.05),与雌激素(倍美力)组比较差异无显著性。结论:健骨二仙丸能有效促进成骨细胞的分化。Objective:Based on the clinical research and animal experiment results,we intend to explore the effect of Jiangu Er xian Pill (JEP) on the differentiation of osteoblast-like cells.Methods:Human osteoblast-like cells were employed,and they were isolated and cultured according to the references.The differentiation of human osteoblast-like cells was observed by the activity of alkaline phos phatase (ALP) and osteocalcin,the number of mineral nodules,and expressing of procollogen 1 α mRNA.The method of PNPP, r...
