补肾活血泄浊汤有效成分对大鼠系膜细胞TGF-βmRNA表达的影响

摘　　要：目的:研究补肾活血泄浊汤有效成分对体外培养大鼠系膜细胞 TGF-β基因表达的影响,以探讨补肾活血泄浊汤治疗肾小球肾炎的机制。方法:应用20%胎牛血清刺激系膜细胞,分别加入补肾活血泄浊汤有效成分川芎嗪、大黄素、灵芝多糖,应用 Northern 杂交技术,观察上述几种中药有效成分对大鼠系膜细胞 TGF-β基因表达的影响。结果:川芎嗪、大黄素、灵芝多糖均可抑制 TGF-β基因的表达,其中以川芎嗪作用最为显著。结论:补肾活血泄浊汤可能通过抑制 TGF-β基因的表达而减轻肾小球硬化。Objective:To investigate the effect of some active principles of Bushen Huoxue Xiezhuo Decoction (BHXD) on expression of transforming growth factor-β mRNA of cultured mesangial rat cells to explore the mechanism by which these active principles sup- pressed the glomerulonephritis.Methods:20 % fetal calf serum as stimulating factor,effects of emodin,Ligustrazine and Ganoderma polysaccharide on TGF-β mRNA expression of mesangial cells were investigated by using the technique of Northern blotting.Results: Emod...
