摘　　要：目的研究血浆置换对肝细胞凋亡的影响。方法应用慢性重型肝炎患者血浆置换治疗前血浆(SHP)、治疗后血浆(SHP')及健康人血浆(P健)、HBV携带者血浆(PHBV)干预HepG2细胞,通过MTT法检测细胞增殖活性,流式细胞术(FCM)检测细胞凋亡,半定量RT-PCR测定凋亡基因FasmRNA的转录水平。结果SHP组HepG2细胞活性显著下降、细胞凋亡严重;SHP'组HepG2细胞活性亦下降,但较SHP组为轻(P<0.01或P<0.05),FCM提示凋亡率显著下降,RT-PCR半定量检测FasmRNA水平显著降低(P<0.01);HBV携带者血浆与健康人比较,对HepG2细胞生长的影响差异无统计学意义(P>0.05)。结论血浆置换治疗可下调Fas的基因转录水平,减轻其凋亡。Objective To investigate the effection of plasma exchange on apoptosis of HepG2 cells.Methods HepG2 cells were cultured with the chronic severe hepatitis patients plasma (before PE called SHP, after called SHP ) , HBV-carriers plasma/serum, well-beings plasma/serum respectively, the proliferation activities of HepG2 cells were detected by MTT method, the apoptosis rates were detected by flow cytometry,the levels of Fas mRNA were detected by semi-quantitative RT-PCR.Results The cytoactive of HepG2 in SHP was...
