白藜芦醇甙对大鼠腹腔巨噬细胞产生TNF-α、IL-6和IL-1及细胞内钙浓度的影响

摘　　要：目的:研究白藜芦醇甙(polydatin,pol)对大鼠腹腔巨噬细胞(peritoneal macrophage,PM)分泌TNF-α、IL-6和 IL-1的作用及其与[Ca^(2+)]_i 的关系,以了解 pol 的抗炎作用机制。方法:采用四甲基偶氮唑蓝[3-(4,5-dimethylthiazol-2-yl)-2,5-diphenylterazolium bromide,MTT]比色法检测 IL-1、TNF-α和 IL-6,采用荧光指示剂和荧光分光光度计检测 PM 的[Ca^(2+)]_i。结果:(1)pol 增加正常大鼠 PM 分泌 TNF-α和 IL-6,对 IL-1的分泌无明显作用。pol 可降低经脂多糖(lipopolysaccharide,LPS)刺激的大鼠 PM 分泌 TNF-α、IL-6和 IL-1。(2)pol 降低正常的和经 LPS 刺激后的大鼠 PM 静息[Ca^(2+)]_i 和外加 CaCl_2后[Ca^(2+)]_i 的升高。结论:pol 对PM 分泌细胞因子有双向调节作用。pol 可以抑制细胞内贮存钙释放和细胞外钙的进入。Pol 的抗炎机制可能与其抑制上述炎症介质有关,并可能通过对炎细胞[Ca^(2+)]_i 的调节发挥作用。Objective:To study the effects of polydatin(pol)on the production of interleukin-1(IL-1),tumor necrosis factor-α and IL-6 se- creted by peritoneal macrophages(PM)and the intracdlular calcium concentration of PM in rats.Methods:The methyl thiazolyl tetra- zolium(MTT)assay was used to measure the IL-1,TNF-α and IL-6.The fluorescence indicator and fluorescence spectrophotometry was used to determine the intracellular calcium concentration to understand the anti-inflammatory mechanism of pol.Results:(1)Com- par...
