姜黄素对四氯化碳诱导大鼠肝纤维化及肝组织Collagen Ⅰ表达的影响

摘　　要：目的:观察姜黄素(Cur)对四氯化碳诱导大鼠肝纤维化及对其肝组织Ⅰ型胶原表达的影响。方法:采用皮下注射40%CCl4复制模型,造模后给予Cur治疗,观察肝组织病理改变并进行肝纤维化分级,并进行血清生化学检测;免疫组化法测定肝组织中CollagenⅠ的变化;RT-PCR检测肝组织中Collagenα1(Ⅰ)mRNA表达。结果:Cur各治疗组大鼠肝纤维化程度较模型组显著减轻;大鼠血清ALT、AST、ALP水平明显降低,A/G比值提高;CollagenⅠ及Collagenα1(Ⅰ)mRNA表达较模型组显著下降。结论:Cur能明显改善肝组织的纤维化程度,提示抑制CollagenⅠ表达是其抗肝纤维化的机制之一。Objective:To investigate the effects of curcumin on expression of collagen Ⅰ in the fibrotic liver induced by carbon tetrachoride in rats.Methods:The liver fibrosis model induced by CCl4 was established in rats and they were treatment with curcumin.Liver fibrosis grades were evaluated on the liver tissue sections and biochemical eaxamination were performed.Immunohistochemistry were performed to detect the expression of collagen Ⅰand reverse transcriptase polymerase chain reaction(RT-PCR)were performed to de...
