夏枯草提取物体内外对小鼠T淋巴瘤EL-4细胞生长的影响

摘　　要：目的:观察夏枯草提取物体内外对小鼠T淋巴瘤EL-4细胞生长的影响,探讨夏枯草提取物的抗肿瘤作用机制。方法:用MTT法、DNA琼脂糖凝胶电泳、TUNEL等方法检测夏枯草提取物体外对小鼠T淋巴瘤EL-4细胞生长的影响。建立C57BL/6小鼠EL-4细胞移植瘤模型,分别给予500、300、100mg/(kg·d)夏枯草提取物,10d后,计算抑瘤率,对肿瘤组织进行形态学观察,并测定其中Bcl-2和Bax蛋白的表达情况。结果:夏枯草提取物体外能抑制EL-4细胞生长,半数抑制浓度(IC50)为(23.67±3.05)mg/L;夏枯草提取物能诱导EL-4细胞凋亡。夏枯草提取物能抑制EL-4细胞在C57BL/6小鼠体内的生长,500、300、100mg/(kg·d)剂量组的抑瘤率分别为(39.54±1.83)%、(65.26±1.31)%、(22.30±3.7)%,平均生存时间分别为(25.50±1.87)、(28.50±2.62)、(24.86±2.19)d,均大于空白对照组(P<0.05),移植瘤Bcl-2蛋白表达降低,Bax蛋白表达增加。结论:夏枯草提取物在体内外均能抑制EL-4细胞的生长,诱导凋亡可能是其抗肿瘤的主要机制之一。Aim:To observe the effects of the extract of Prunella vulgaris L on the growth of mouse T lymphoma cells EL-4 in vivo and in vitro,and explore its mechanism of anti-tumor.Methods:The growth inhibition of EL-4 cells cultured with the extract of Prunella vulgaris L was measured by MTT assay,and the apoptosis was detected using agarose gel electrophoresis,TUNEL.The anti-tumor activity of the extract of Prunella vulgaris L in the experimental EL-2 transplantation tumor in C57BL/6 mouse was evaluated.Results:The...
