高效液相色谱法测定β-氨基醇类药物中间体的含量及ee值

摘　　要：目的:建立高效液相色谱法测定生物转化液中2种β-氨基醇类手性药物中间体含量及ee值的方法。方法:以Chiralcel OJ-H为手性分离柱,正己烷-异丙醇(85:15)为流动相,检测波长273nm,进样量20μL。生物转化液用乙酸乙酯萃取后进行测定。结果:2种中间体的分离效果良好,2种中间体在0.25～1.25g/L范围内线性关系良好,它们的平均加样回收率分别为98.18%和98.86%。在测定中间体含量的同时,还能测定样品的ee值。结论:该方法准确灵敏,精密度好,适合于生物转化液中β-氨基醇类手性药物中间体的测定。Aim:To establish an HPLC method to determine the ee values and content of two β-aminoalcohol chiral drug intermediates in bioconversion broth.Methods:The determination was performed on a Chiralcel OJ-H column with a 85:15 mixture of n-hexane and isopropanol alcohol as mobile phase,determination wavelength of 273 nm and injection volume of 20 μL.
