摘  要:目的 为进一步研究脑内神经源型NO合酶 (nNOS)免疫阳性神经元的表达提供可靠的实验方法。方法 ABC免疫细胞化学方法。结果 大鼠大脑皮质、尾壳核、海马、中脑、延髓、小脑等中枢神经系统的各个部位均有nNOS免疫阳性神经元和神经末梢出现 ,nNOS免疫阳性神经元胞体呈棕褐色 ,细胞核处颜色浅淡 ,突起有一个或多个 ;nNOS免疫阳性神经纤维呈棕色串珠状 ,且交错分布。结论 ABC法是显示NOS亚型nNOS表达的可靠方法 ,但一抗浓度不宜过高 ,而且需延长抗体孵育的时间。Objective To supply a credible experimental method for study the expression of nNOS-immunoreactive neurons in rat brain. Methods The ABC immunocytochemistic technique was used in brain tissues of 12 rats. Results The nNOS-immunoreactive neurons and innervations appeared in the rat brain, such as cerebral cortex, caudate putamen, hippocampus, midbrain, medullaery bulbus and cerebellum. The nNOS-immunoreactive neurons was brown, the color of nucleus was weak and the bodies of nNOS-immunoreactive neurons had one or more protuberances. The nNOS-immunoreactive innervations exhibited brown strings beadings and had an interlacing distribution. Conclusions The ABC method is credible for study of the expression of nNOS-immunoreactive neurons, at the same time, the moderate concentration of the first antibody is advisable and the time of incubation should be prolonged.
