摘  要:目的 :建立促红细胞生成素 ( EPO)的放免分析方法 ,并探讨血清 EPO浓度测定的临床意义。 方法 :用rh EPO免疫动物制备了高质量的抗血清 ,建立了 EPO放免分析方法 ,并测定了正常人和贫血、肾衰患者血清 EPO值。 结果 :灵敏度为 0 .2 2 μg/L,批内 CV为 6.9%~ 8.4% ,批间 CV为 8.0 %~ 9.6% ,回收率为 93 .8%~110 .4%。正常血清 EPO值为 ( 1.5 9± 0 .69) μg/L,贫血患者为 ( 3 .64± 1.2 8) μg/L,肾衰患者为 ( 0 .85± 0 .2 5 ) μg/L,与正常人之间有显著性差异 ( P<0 .0 0 1)。 结论 :EPO的放免分析方法是一种简单、灵敏、特异的分析方法 ,可满足临床的需要 ,测定血清Objectives: To develop a method of radioimmunoassay for erythropoietin(EPO) determination and to explore its clinical value.  Methods: ①Anti-recombinant EPO antibody with high affinity was prepared to develop the radioimmunoassay of EPO;②Serums EPO levels were measured in normal subjects and in patients with anemia and renal failure. Results: The sensitivity was 0.22 μg/L,intra- and inter-assay variation were 8.0%~9.6%、6.9%~8.4%, respectively. Serums EPO levels in normal subjects and patients with anemia and renal failure were (1.59±0.69)μg/L、(3.64±1.28)μg/L and (0.85±0.25)μg/L, respectively; There were significant differences in serum concentrations of EPO among normal subjects and patients with anemia and renal failure (P<0.001).  Conclusions: This radioimmunoassay of EPO fulfills reasonable requirements of a clinical routine in terms of specificity, sensitivity and practicability. Measurement of EPO concentration is useful for clinical diagnosis and choice of treatment of anemia and renal failure.
