摘  要:目的 研究GnRH类似物(阿拉瑞林)诱导胃平滑肌细胞内钙动员的机制。方法 采用共聚焦技术检测细胞内钙荧光强度的变化。结果 (1)在Hanks液中,GnRH类似物浓度为107、106、10-5mol-L.-1时,胃平滑肌细胞内Ca2+逐渐升高,其峰高(peak-resting)分别为 6.00 ± 0.50、9.23± 0.62、18.97± 2.42,其达峰时间分别为 42.37± 1.65、54.93± 3.22、105.63 ± 3.92s,井呈时间和剂量依赖性;(2)当阿拉瑞林浓度为10-4mol·L-1时,胞内Ca2+略有升高,与10-5mol·L-1相比(P<0.01);(3)在有外钙和无外钙时,阿拉瑞林对胞内Ca2+的升高亦有显著差异(P<0.01);(4)在无外钙并加入内罗陡孵育后,GnRH引起的胞内 Ca2+轻度升高,但与 D-Hanks组相比无显著性差异(P<0.05);(5)在 Hanks液中,用拉西地平孵育后,再加入阿拉瑞林,其胞内Ca2+仍可升高,井有显著差异(P<0.01)。结论Ca2+参与了GnRH类似物阿拉瑞林对胃平滑肌细胞作用的细胞内分子转导过程。细胞内Ca2+的增加既源于内钙的释放又源于外?Objedctive To study the mechanism of GnRH agal ogus( Alarelin)on[Ca2+]i mobilization in stomach smooth muscle cells (SSMC)o f rats.Methods Cells were cutured and loaded with Fluo -3-AM.[Ca2+]i was measured by fluorescent intensity(FL)in each cell with confocal microscopy.Results (1)In Hanks solution,10 - 7,10-6,10 -5mol*L-1Alarelin can elevate[Ca2+]i , its peak-resting values reached 6.00±0.50、9.23±0.62、18.97±2.42 respec tively,which indicate that the level of [Ca2+]i act in an dose-depen de nt and time-dependent.(2) Contrarily,when Alarelin was 10-4mol*L-1 , its peak-resting value only reached 6.32±0.67, conpared with Alarelin 10 -5mol*L-1,which was sigificantly lower(P<0.01.(3)[ Ca2+]i wa s also singificantly higher in Hanks solution than that in D-Hanks solution(P< 0.05).(4)In D-Hanks solution with ryanodine,Alarelin had no effect on levels of [Ca2+]i,compared with group of D-Hanks(P>0.05).( 5)When pertreatment with Lacidipine in Hanks solution,the effect of Alarelin 10-5mol*L -1 was partly inhibited(P<0.01)。Conclusion The data indicate that[Ca2+]i plays an important role in molecular mechanism of intrace llular singal transduction pathways which GnRH analogue,Alarelin mediate in SSM C .T he increasing of[Ca2+]i was caused by releasing from intracellular sto r es and extracellular Ca2+influx,possibly through L-type calcium channels .
