摘 要:牙周炎和种植体周围炎是牙菌斑引起的发生在牙周支持组织上的慢性感染性炎症,通常可导致牙周组织结构破坏,引起牙和种植体松动脱落。课题组前期研究发现中药肉桂能杀灭牙周可疑致病菌并具有良好的抗炎作用。目的:桂皮醛对成骨细胞的增殖及成骨功能是否具有影响。方法:采用MTT法,测定五种浓度的药物对成骨细胞生长的抑制率,同时,通过对碱性磷酸酶活力、钙含量的测定、茜素红染色显示矿化结节等实验方法考察不同浓度的药物组对成骨细胞功能的影响。结果:1.低浓度的桂皮醛提取液8mg/L,16mg/L组同空白组相对照,其细胞增值水平较高,而在高浓度如128mg/L,64mg/L,32mg/L的稀释比时,成骨细胞的增值则出现一定程度的减弱。2.桂皮醛提取液在稀释比为8mg/L、16mg/L时,与对照组相比,成骨细胞的ALP活性及钙含量有显著增加,成骨细胞的矿化结节数有少量的增加(p<0.05),而128mg/L、64mg/L、32mg/L的高浓度组,成骨细胞的ALP活性及钙含量则有不同程度的降低,矿化结节数减少(p<0.01)。结论:中低浓度(16mg/L、8mg/L)的桂皮醛对成骨细胞具有一定的正向调节作用,高浓度(128mg/L、64mg/L32mg/L)的桂皮醛提取液对成骨细胞具有一定的细胞抑制作用,对成骨细胞的增值、分化、成熟及功能有不同程度的影响。Periodontitis and Peri-implantitis are chornic infectional inflammation that emerges on the periodontium and infected by dental plaque.It usually induces the disorganization of periodontal tissue,which will eventually develop into attachment loss with the defluxion and lossening of tooth,as is the main cause of losing tooth for adults.Objective :To investigate the effect of Cinnamic aldehyde(CA)on growth of Osteoblasts.Methods:The MG63 cells were inoculated and exposed to different concentrations of CA(128,64,32,16,8 ugml-1 respectively).The proliferation rates were assessed by an MTT assay.MG63 cell differentiation function was accessed by Alkaline phosphatase(ALP)activity.Results:1.The proliferation of MG63 cells with 16、8ugml-1 CA had no significant difference compared with controls.16、8ugml-1 CA obviously promoted the ALP activity of MG63 cells.CA(128,64 and 32ugml-1)remarkably inhibited the proliferation and the ALP activity of MG63 cells.2.16mg/L,8mg/L diluted than in the role of liquid cinnamaldehyde,with the control group,osteoblast mineralization nodules for a small increase(p <0.05).Conclusion:low concentration(l6mg/L 8mg/L)CA on osteoblasts have some positive effect,a high concentration(128mg/L,Fas 32g)CA extract of osteoblast cells with a certain effect,right osteoblasts value,differentiation,maturation and function of varying degrees of impact.
