Effects of Production Performance,Eggshell Quality and Metabolism Modulation by Injecting pCEP4-PTH Plasmid into Layers

摘  要:试验选用56周龄新扬州鸡产蛋鸡150只,随机分为3个处理,每个处理5个重复,各重复10只。研究经腿肌注射0,200,400μg鸡甲状旁腺素(pCEP4-PTH)基因质粒对蛋鸡生产性能、蛋壳质量、血液部分激素和代谢物浓度的影响。结果表明:pCEP4-PTH基因质粒能提高蛋鸡产蛋率,与对照组相比,200μg组和400μg组分别提高4.70%(P<0.05)、5.83%(P<0.05),400μg组料蛋比显著降低(P<0.05),200μg组破软壳蛋率显著低于对照组(P<0.05);200μg注射量可显著改善蛋比重、蛋壳强度和蛋壳比例(P<0.05),400μg注射量对蛋壳质量各参数影响不显著(P>0.05);各组间血钙、血磷及碱性磷酸酶差异不显著(P>0.05);200μg和400μg组甲状旁腺素(PTH)水平均显著升高(P<0.05),且注射400μg时达到极显著水平(P<0.01),注射400μg pCEP4-PTH基因质粒时降钙素(CT)和雌二醇(E2)显著升高(P<0.05);两试验组骨钙素(BGP)与对照组间差异不显著(P>0.05)。蛋鸡肌肉注射200μg pCEP4-PTH质粒生产性能和蛋壳质量最佳。One hundred and fifty 56-week-old New Yangzhou layers were randomly divided into three groups,each group had five repeats,and each repeat had 10 birds.Effects of injecting pCEP4-PTH gene plasmid into the crural muscle of layers with the dose of 0μg,200 μg and 400 μg on production performance,eggshell quality,serum hormone and metabolites levels were studied.The results showed that pCEP4-PTH gene plasmid enhanced the laying rate by 4.70%(P<0.05),5.83%(P<0.05) in 200 μg treatment and 400 μg treatment,and feed/egg ratio significantly decreased in 400 μg treatment(P<0.05),soft-shell egg ratio in 200 μg treatment was significantly lower than that of the control group(P<0.05);injecting dose of 200 μg might obviously improve egg proportion,eggshell strength and eggshell weight percentage(P<0.05),injecting 400 μg dose might not affect eggshell quality parameters obviously(P>0.05);there was no difference in blood calcium,phosphours and AKP among three treatments(P>0.05);in 200 μg and 400 μg treatments,the level of PTH increased significantly(P<0.05),and in 400 μg dose it increased extremely significantly(P<0.01);injecting dose of 400 μg could increase CT and E2 level(P<0.05),BGP in the two treatments was not significant compared with the control group(P>0.05).Production performance and the eggshell quality were the best when injecting dose was 200 μg pCEP4-PTH plasmid.
