摘  要:目的观察姜黄素对单纯性肥胖大鼠胰岛素抵抗的影响,并研究其作用机制。方法健康雄性SD大鼠40只,随机分为正常组、模型组、姜黄素高剂量组和姜黄素低剂量组,每组10只:除正常组喂食普通饲料外,其余各组喂食高脂饲料4周诱发肥胖致胰岛素抵抗模型,建立单纯性肥胖大鼠模型。姜黄素干预组再给予低、高剂量姜黄素灌胃4周,实验结束后采血测定血清三酰甘油(TG)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)、游离脂肪酸(FFA)、血糖(GLU)及血胰岛素(INS)并计算胰岛素敏感指数(ISI),剖杀动物测睾周和肾周体脂/体质量。结果(1)模型组空腹GLU、INS、TG、LDL和FFA与对照组比较差异有统计学意义(P<0.05),而HDL及ISI显著高于对照组;(2)姜黄素低、高剂量组空腹GLU、INS、TG、LDL和FFA低于模型组,同时姜黄素低、高剂量组HDL及ISI与模型组相比,模型组显著升高,姜黄素高剂量组尤为明显;(3)模型组体脂/体质量与对照组比较,差异有统计学意义(P<0.05)。经姜黄素干预后低、高剂量组体脂/体质量与模型组比较,差异均有统计学意义(P<0.05)。结论姜黄素能够降低单纯性肥胖大鼠体质量及体脂,且具有降低血糖、调节血脂、改善胰岛素抵抗的作用。Objective To observe the effect of curcumin on insulin resistance in simple obesity rats and to discuss the possible mechanism.Methods Forty healthy male SD rats were randomized into groups of normal control,model,high-dosage curcumin(HDC group) and low-dosage curcumin(LDC group),10 in each.The control group were fed with ordinary diet,while the other 3 with high-fat diet,4 weeks,to induce obesity-caused insulin resistance models,to establish simple obesity models.Besides,curcumin intervention groups were given low- or high-dosage curcumin,4 weeks,and after experiment triacylglycerol(TG),high density lipoprotein(HDL),low density lipoprotein(LDL),free fatty acids(FFA),glucose(GLU) and insulin were determined,insulin sensitivity index(ISI) were calculated and then the animals were sacrificed to detect the body fat /body mass of kidney and testicles.Results Model group were significantly different from control in fasting GLU,insulin,TG,LDL and FFA(P<0.05),and their HDL and ISI were significantly higher than those of control.Fasting GLU,INS,TG,LDL and FFA of groups LDC and HDC were lower than those of model group,and HDL and ISI of model group significantly increased as compared with groups LDC,HDC,especially with HDC.There was significant difference in body fat/body mass between model group and control(P<0.05).Groups LDC and HDC were significantly different from model group in body fat/body mass after curcumin intervention(P<0.05).Conclusion Curcumin can inhibit weight gain caused by high-fat diet,reduce the levels of TG,LDL,FFA,glucose and insulin,and improve insulin resistance and metabolic disorders.
