摘  要:目的 建立一种简便的大鼠前脂肪细胞的原代培养方法.方法 无菌取成年Wistar大鼠的腹股沟纯脂肪颗粒,采用酶消化法进行原代培养.在倒置显微镜下观察细胞形态及生长情况,以油红O脂肪染色法进行原代培养细胞的鉴定.结果 培养细胞的形态学观察显示,脂肪细胞贴壁后初为类圆形,3 d后渐成梭形;7~8 d细胞增殖明显,形状由多角梭形变为椭圆形,并开始积聚脂肪颗粒;10 d后细胞呈椭圆形、圆形,胞内积聚大量脂肪颗粒.培养10 d的细胞经油红O染色后于普通光镜下观察,见细胞内出现红染颗粒,可以鉴定细胞分化为前脂肪细胞.结论 成功建立了大鼠前脂肪细胞的原代培养方法,为脂肪内分泌学研究提供了制备适用细胞模型的手段.Objective To establish a new method for primary culture of rat preadipocytes. Methods Using enzyme-digesting method,fibroblast-like cells from the rat adipose tissues were cultured. The morphological changes of the cultured cells were observed under an inverted microscope and stained with oil red O to identify the primary cultured cells. Results Morphologically,the attached fat cells were round-like in shape initially,and then changed into spindle after 3 days. On day 7 to 8,the cells proliferated actively,and the polygonal/spindle cells turned into oval cells with fat particles accumulated inside. On day 10,the cells were oval of round in shape with a large amount of fat particles in the plasma. After being cultured for 10 days,red-stained particles were observed in the oil red O-stained cells under a light microscope,indicating that the cells had differentiated into preadipocytes. Conclution We have successfully established a method for primary culture of rat preadipocytes,which provides a basis to establish appropriate cell models for studies on adipose endocrinology.
