摘  要:目的观察体外培养条件下葡萄糖浓度对3T3-L1脂肪细胞诱导分化过程中UCP2mRNA表达水平的变化。方法体外培养3T3-L1脂肪细胞,在诱导3T3-L1脂肪细胞分化成熟的基础上,经不同葡萄糖浓度5.5,25,45mmol/L培养后,3T3-L1脂肪细胞于第3,5,7天抽提总RNA,用半定量逆转录聚合酶链反应(RT-PCR)检测UCP2mRNA的表达。结果随着脂肪细胞逐渐分化成熟,在第7天,45mmol/L葡萄糖终浓度培养条件下,3T3-L1脂肪细胞UCP2mRNA的表达受到抑制(P<0.05);5.5mmol/L葡萄糖终浓度培养条件下,3T3-L1脂肪细胞可增加UCP2mRNA的表达(P<0.01)。结论体外培养中,葡萄糖浓度的变化对脂肪细胞中UCP2mRNA的表达与调控具有一定的影响。Objective To investigate the in vitro effect of glucose concentration on UCP2 mRNA expression in 3T3-L1 adipocytes. Methods The differentiated 3T3-L1 adipocytes were incubated with 5.5,25,45mmol/L glucose respectively for 3 days,5 days and 7 days. The total cellular RNA was extracted, and the expression of UCP2 mRNA were detected by RT-PCR. Results High concentration of glucose (45mmol/L) suppressed the expression of UCP2 mRNA (P<0.05), and low concentration of glucose(5.5mmol/L) increased the expression of UCP2 mRNA(P<0.01),both compared with a control group (25mmol/L). Conclusion In vitro studies,the concentration of glucose is a very impact factor for the regulation of UCP2 mRNA expression.
