摘  要:目的采用单细胞凝胶电泳法研究双酚A对小鼠睾丸细胞DNA的影响。方法制备小鼠生殖细胞悬液,用不同浓度的双酚A染毒1 h后,应用单细胞凝胶电泳法检测睾丸细胞DNA损伤情况。结果与对照组比较,除10-10mol/L组外,其他各染毒组拖尾率均明显高于对照组(P<0.05)。随着双酚A染毒浓度的增加,小鼠睾丸细胞DNA损伤程度也逐渐增加(P<0.05),在10-5mol/L和10-6mol/L组均出现了Ⅳ级损伤。结论双酚A体外染毒可造成小鼠睾丸细胞DNA损伤,且随着剂量的增加有加重的趋势。Objective To study the DNA damage to male mice testis cells induced by BPA with single cell gel electrophoresis.Methods Mice Testis cell suspension was prepared and infected with different dosage of BPA.An hour later,DNA damage to mice testis cells was measured with single cell gel electrophoresis.Results The ratios of tail tailings in different dosage of BPA groups were significantly higher than that in the negative control group(P<0.05) except the 10-10 mol/L group.The DNA damage level increased with the increase of the BPA dosage(P<0.05).Ⅳ grade DNA damage was found in the 10-5 and 10-6 mol/L group.Conclusion BPA could induce DNA damage in isolated male mice testis cells.The degree of DNA damage was aggravated with the increase of the BPA dosage.
