摘 要:目的比较胶原川芎嗪缓释剂穴位埋药与腹腔注射川芎嗪对脑缺血再灌注后大鼠海马炎性细胞因子TNF-α含量的影响,以期为针灸临床治疗提供更有效的方法。方法将SD大鼠随机分为正常组、假手术组、模型组(24h组、72h组、120h组)、穴位埋药组(24h组、72h组、120h组)、腹腔注射组(24h组、72h组、120h组)。采用改良线栓法制备局灶型脑缺血(MCAO)再灌模型,以胶原川芎嗪制备的缓释剂(药线)在大椎、双侧内关穴位埋药。用放射免疫法观察各组大鼠海马组织中TNF-α含量的变化。结果脑缺血再灌注模型24h组大鼠海马组织TNF-α含量即明显升高达到峰值,之后逐渐下降,与正常组,假手术组比较有显著性差异(P<0.01);治疗各组大鼠缺血侧海马组织TNF-α含量均有所降低,与模型组同时相比较有显著意义(P<0.01),并随时间的推移逐渐趋于正常。穴位埋药组治疗效果明显优于腹腔注射组(P<0.01)。结论穴位埋药可降低脑缺血再灌模型大鼠海马组织TNF-α的水平,减少炎症反应引起继发的脑神经元损伤,疗效优于腹腔注射组。Objective To observe the effects of embedding implantation of collagen and tetramethylpyrazine in points and injecting tetramethylpyrazine into abdomen on the content of TNF-α of cytokine in hippocampus of rat with focal cerebral ischemia reperfusion.Methods SD rats were randomly divided into normal group,sham operation group,model 24h group,model 72h group,model 120h group,injecting abdomen 24h group,injecting abdomen 72h group,injecting abdomen 120h group,embedding medicine in points 24h group,embedding medicine in points 72h group,embedding medicine in points 120h group.Model of local ischemia was used by middle cerebral artery occlusion(MCAO),preparation of implantation collagen and tetramethylpyrazine to embed the Points of Dazhui and both Neiguan,detected the Content of TNF-α in hippocampus by immunohistochemistry and radio immunoassay(RIA).Results After ischemia 30min and reperfusion 24h,the content of TNF-α in hippocampus of model group rats reached peak value and then dropped gradually,which was significantly higher than those of normal and sham operation group rats(P<0.01).Compared with model group,the content of TNF-α in hippocampus of each treatment group was obviously lower(P<0.01)and then decreased with the lapse of time.The effect of embedding implantation of collagen and tetramethylpyrazine in points was excelled the group of injecting abdomen distinctly(P<0.01).Conclusion Embedding implantation of collagen and tetramethylpyrazine in points can decrease the content of TNF-α in hippocampus of rat brain after cerebral ischemia reperfusion,cut down neuronal cells damnification by inflammation reaction,and has protection effects on neuronal cells after cerebral ischemia reperfusion in rats.
