补骨脂对体外培养新生大鼠颅骨成骨细胞的影响

摘　　要：目的观察由补骨脂乙醇提取物中分离得到的单一化学成分对体外培养新生大鼠颅骨成骨细胞增殖、分化的影响。方法在新生大鼠颅骨成骨细胞体系中加入不同化合物,检测细胞内碱性磷酸酶含量,并用MTT法检测细胞增殖活性。结果由补骨脂乙醇提取物中分离出的化合物Ⅰ、Ⅱ、Ⅷ、Ⅸ、Ⅹ能提高大鼠颅骨成骨细胞ALP活性及促进其增殖。结论化合物Ⅰ、Ⅱ、Ⅷ、Ⅸ、Ⅹ可促进成骨细胞分化、增殖,为补骨脂治疗骨质疏松的有效成分。Objective To investigate the effect of the simple components separated from Psoralen Corylifolia L.on the proliferation and differentiation in neonatal rat calvarium.Methods New born SD rat calvarium osteoblasts were cultured in DMEM alone or exposed to different componets of Psoralen Corylifolia L.The osteoblast differentiations were evaluated by detecting activities of alkaline phosphatase (ALP).The MTT method was employed to detect the osteoblast proliferation after culture with different components.Results The componentsⅠ,Ⅱ,Ⅷ,Ⅸ,Ⅹ separated from Psoralen Corylifolia L. increased ALP activity and proliferation of rat cultured osteoblasts.Conclusion The components Ⅰ,Ⅱ,Ⅷ,Ⅸ,Ⅹ can stimulate the differentiation and proliferation of neonatal rat calvarium osteoblasts in vitro and may be effective components in Psoralen Corylifolia L. which used to treat osteoporosis.
