摘 要:目的通过实验明确大鼠口服LMWH的绝对生物利用度,为研制LMWH口服制剂提供依据。方法从大鼠的肝静脉采血,应用柱层析-比色法测定血中LMWH的含量,求得LMWH的绝对生物利用度。结果测定方法的标准曲线方程是A=0.515 8 C-0.0006(r=0.999 8,n=6),回收率是97.14%(RSD=1.03%,n=5),日内、日间标准差分别是6.8%、6.4%。求得绝对生物利用度89.61%。结论本实验证实,研制LMWH口服制剂是可行的。Objective To provide significant basis for development of LMWH oral preparation though determination of absolute bioavailability of rats after oral LMWH.Methods Blood was sampled through hepatic vein in rats,and the absolute bioavailability of LMWH can be calculated based on its concentration in blood, which was determinate by column chromatogram-colorimetry.Results The equation of calibration curve was A =0.5158C-0.0006 with a correlation coefficient of 0.9998.The recovery was 97.14% and RSD was 1.03%(n =5).Within-day and between-day RSDs were 6.8% and 6.4%,respectively.The absolute bioavailability was 89.61%.Conclusion The result shows that development of LMWH oral preparation is feasible.
