摘  要:目的观察脑源性神经营养因子(BDNF)对于烧伤大鼠血清造成基底神经元损伤的保护作用。方法取出生后1d乳鼠前脑基底神经元培养,随机分为正常对照组、损伤组(烧伤大鼠血清损伤组)、BDNF保护组。测定烧伤大鼠基底神经元NO和LDH含量;给予原代培养基底神经元不同浓度BD-NF24h后,加入不同浓度烧伤大鼠血清,测定细胞存活率和培养液中NO含量。结果大鼠烧伤后基底组织NO和LDH含量明升高,烧伤大鼠血清可引起培养的基底神经元存活率下降,培养液中NO含量升高;BDNF能降低基底组织中NO和LDH的含量,提高培养的基底神经元的存活率,减少培养液中NO含量,其作用呈剂量依赖性,BDNF对神经元存话率的影响与NO含量呈显著负相关。结论BDNF对烧伤大鼠血清引起的基底神经元损伤有保护作用,其作用机制可能是通过抑制NO的神经毒性。Objective To investigate the protective effects of brain-derived neurotrophic factor(BDNF) on primary cultured dissociated basal ganglia neurons which were mediated by serum of burn.Methods Basal ganglia cultures were made of 1-day-old rats.The slices were divided into 3 groups,control group,injury group (serum of burn group) and BDNF group.The contents of NO and LDH of striatum were measured.Using primary cultures of rat striatal neurons,the effects of BDNF on the neurons survival and the content of NO which was induced by serum of burn rat were observed.Results The contents of NO and LDH increase in burn rats.The serum of burn rat could decease the cell survival rate and increase the levels of NO and BDNF could promote neurons survival,deceased NO level which induced by serum of burn rat.The protective effects of BDNF against cytotoxicity were dependent on concentrations.Conclusion BDNF protect basal ganglia neurons from the injury by serum of burn rat,the mechanism was related to prevent NO neurototoxicity.
