早期短期应用17β-雌二醇抑制兔颈动脉球囊损伤后新生内膜增殖

摘  要:目的探讨短期应用17β-雌二醇对血管球囊损伤后新生内膜增殖的影响。方法将颈总动脉球囊损伤的去卵巢兔分为10组,在不同时间点给予17β- 雌二醇(20μg/kg/d)。分组:①损伤前三天开始给药,持续到术后第三天;②损伤前三天开始给药,持续到术后第七天;③损伤前三天开始给药,持续到术后第十四天;④手术当天开始给药,持续到术后第三天;⑤手术后第七天开始给药,持续到术后第十四天;⑥损伤后每天给予棉花油;⑦仅在损伤前一天给药一次;⑧仅在损伤前15min给药一次;⑨仅在损伤后第一天给药一次;⑩在损伤后第一天开始给药,连续2d。颈总动脉球囊损伤两周后处死所有动物,取损伤的颈总动脉,进行血管形态学分析。结果术后15min应用雌激素,连续应用3d,可使新生内膜面积减少59%(P<0.05,vs vehicle),其效果与从手术前三天开始应用,且持续用到术后十四天相同(61%,P>0.05,vs 0~3d);从术前三天开始应用雌激素,持续用到术后第七天,不能增强雌激素的抑制作用(P>0.05,vs 0~3d);从术后第七天开始给药,持续到第十四天,雌激素的抑制作用不明显(P>0.05,vs vehicle);仅在术后第一天或第二天应用雌激素,也可抑制新生内膜的形成(P<0.05,vs vehi- cle);在术前或手术当天应用雌激素,没有效果(P>0.05,vs vehicle)。结论 17β- 雌三醇抑制受损血管新生内膜的形成,其作用的发挥主要在血管损伤后72h内。Objective The goal of this investigation was to assess the effects of Short - Term 17β-estradiol treatment on the neointimal proliferation after Balloon Injury. Methods The animal underwent ovariotomy and balloon injured were divided 10 blocks, then give the Short- term 17β-estradiol(20μg/kg/day) in deferent time: (1)from the 3rd day before balloon injured to the 3rd day after operated (2)the 3rd day before balloon injured to the 7th day after operated (3)the 3rd day before balloon injured to the 14th day after operated (4)the day operated to the 3rd day after (5)the 7th day to the 14th day after operated (6)vehicle 100ml cottonseed oil SC daily (7)a single SC injection (20μg/kg/day) 1st day before balloon injured (8) a single SC injection (20μg/kg/day) 15 minutes before balloon injured (9) a single SC injection (20μg/kg/day) 1st day after balloon injured (10) SC injection 2 days after balloon injured. Results 17β-estradiol treatment initiated 15 minutes before injury and continued for only 3 days after injured was associated with a comparable 59% (P<0.05 vs Vehicle) reduction in neointima formation. Neointima formation was reducedby 61%(P<0.05 vs Vehicle) in animals treated daily from 3rd day before injury until the rabbits were killed 14th day after injury. In contrast, 17β-estradiol treatment initiated at 7TH day after injury and continued until the 14th day had no effect on neointima formation(P > 0.05 vs Vehicle). Conclusion 17β- estradiol inhibits neointima proliferation by inhibiting early vascular injury response that occurred in early 72 hours after balloon injured.
