清肺饮合百令胶囊对哮喘大鼠血清IL-4、IL-5和IFN-γ的影响研究

摘　　要：目的:探讨清肺饮合百令胶囊对哮喘急性发作期大鼠Th1／Th2细胞亚群的影响及防治哮喘的作用机制。方法:将50只SD雄性大鼠随机分为正常组、模型组、清肺饮合百令胶囊组、清肺饮组、地塞米松组,每组10只。除正常组外,以OVA混悬液诱发哮喘,造模第15d后分别给予相对应的药物灌胃,用酶联免疫吸附试验(ELISA)检测血清中白细胞介素-4(IL-4)、白细胞介素-5(IL-5)、干扰素-γ(IFN-γ)的变化。结果:与正常组比较,哮喘模型组大鼠IL-4、IL-5含量显著升高,IFN-γ含量明显降低(P<0．01),IL-4／IFN-γ比例明显失衡。与哮喘模型组比较,各药物治疗组IL-4、IL-5含量显著下降(P<0．01),IFN-γ含量明显升高(P<0．01),IL-4／IFN-γ显著下降(P<0．01),结论:清肺饮合百令胶囊具有调节Th1／Th2的失衡和抑制IL-5的异常增多所导致的嗜酸粒细胞(Eos)趋化和募集的作用。Objective:To study the effects of Qingfei Yin combined with Bailing capsule on Th1/Th2 cell subpopulation and the mechanism of protecting and curing asthma in rats. Methods: Fifty male SD rats were divided randomly into 5 groups, 10 rats per group: normal control, asthmatic model, Qingfei yin/Bailing capsule, Qingfei yin and desamethasone group. Except for normal group, all the asthmatic model rats were induced by OVA suspension and poured with relative medicine after 15 days.The level of IL-4,IL5 and IFN-γin serum were measured by ELISA. Results: Compared to normal group, the content of IL-4 and IL-5 in model group increased significantly while content of IFN-γdecreased significantly(P<0.01), the ratio between IL-4 and IFN-γwas in imbalance. Compared to model group, the content of IL-4 and IL-5 decreased significantly(P<0.01), the content of IFN-γincreased significantly(P<0.01), the ratio between IL-4 and IFN-γdecreased significantly(P<0.01) in all treatment groups. Conclusion Qingfei Yin plus Bailing capsule iseffective to adjust Th1/Th2 imbalance and inhabit abnormal increase of IL-5 resulting in chemotaxis and collection of eosinophil(Eos) in asthmatic rats.
