摘  要:目的:应用现代分子生物学技术获取中药材及其超微饮片的ITS序列,通过分析比较,为超微饮片的质量控制提供可靠方法。方法:以金银花、吴茱萸的干药材和超微饮片为实验材料,提取总DNA,克隆和测定其rDNA的转录间隔区(internal transcribed spacer,ITS)序列。结果:获得了样品中药材的rDNA的ITS1,ITS2,5.8S,18S和28S序列,金银花、吴茱萸的传统饮片和超微饮片的ITS序列的相似度在99%以上。结论:利用ITS序列分析可以科学有效地对超微饮片进行鉴定。Objective:To obtain and compare the ITS sequence from Chinese materia medica with the molecular biological technique, and provide the dependable criterion for the identification of ultramicro capsule from Chinese materia medica. Methods: From the experimental dry materials and ultramicro capsule from Flos Lonicae and Evodia Rutaecarpa, the total DNA was distilled, cloned to detect the internal transcribed spacer(ITS) ultramicro capsule of ribosomal DNA. Results:The sequences of ITS1,ITS2 and 5. 8S. of the two herbals were obtained. Their similitude was more than 99%. Conclusion:The analysis of ITS sequence can provide an effective criterion to identify the ultramicro capsule from Chinese materia medica.
