蜈蚣提取物诱导SiHa细胞凋亡及其作用机制

摘　　要：目的研究蜈蚣提取物对宫颈癌SiHa细胞生长的抑制及其作用机制。方法观察不同浓度的不同蜈蚣提取物对宫颈癌SiHa细胞的生长抑制情况:以不同浓度的蜈蚣提取物和顺铂作用于SiHa细胞48h,MTT法测定药物对细胞生长的影响,流式细胞术观察DNA含量和凋亡的改变,DNA片断化分析凋亡特有DNA梯形格局,Western blot测定bax、Caspase3蛋白水平的表达。结果SiHa细胞用8、16、32mg/mL的乙醚、乙醇蜈蚣提取物处理48h后,细胞生长显著受抑制;DNA周期改变明显,亚G1峰与对照组有显著性差异;DNA-Ladder显著;Western blot提示相关凋亡蛋白bax、Caspase3表达并显示活性,凋亡率增高明显。结论中药蜈蚣乙醚、乙醇提取物在体外对宫颈癌SiHa细胞的生长有明显地抑制作用,机制与改变细胞DNA周期,促进凋亡有关。Objective To study the effects of centipede extracts on proliferation of cervical cancer cell line SiHa and its mechanism.Methods SiHa cell treated with different concentrations of centipede extracts in vitro.The growth of the treated SiHa cell was observed by MTT;DNA cycle and apoptosis of the SiHa cell were detected by flow cytometry.DNA fragmentation assay was used to demonstrate effects of centipede extracts on SiHa cell line.Protein expression levels of bax and caspase 3 were checked by Western blottin...
