摘  要:探讨不同状况下全反式维甲酸对转染IGF—Ⅱ基因胃癌细胞生物学行为的影响。方法以人胃癌细胞系SGC7901细胞分别转染IGF—Ⅱ反义基因.以不同浓度全反式维甲酸处理,观察细胞生长曲线。以MTT法测细胞生长抑制,DAPI染色计数细胞凋亡。扫描及透视电镜观察细胞形态。结果在全反式维甲酸干预后,转染IGF—Ⅱ反义基因细胞凋亡指数上升,且与药物浓度正相关j细胞生长抑制;电镜显示细胞结构改变。结论反义基因IGF—Ⅱ转染胃癌后联合维甲酸可协同抑制胃癌细胞恶性表型,诱导细胞凋亡。The target of our research is to the effect of ATRA on apoptosis of gastric cancer cells transfected with antisense IGF-Ⅱ gene.We transfected antisense IGF-Ⅱ gene into SGC-7901 cell line, and treated with ATRA in a variety of concentration, the rate of apoptosis was detected by DAPI, and we observed the cells entity with scanning electron microscopy and transmission electron microscopy.Apoptosis was detected in antisense IGF-Ⅱ transfected gastric cancer cells treated with ARTA, which was positively related with theconcentration of ATRA.Morphological apoptotic appearance were observed.IGF-Ⅱ gene play an important role in mediating the apoptosis of gastric cancer line induced by ATRA, which is likely the mechanism for ATRA in the growth inhibition of gastric cancer cell.
