摘 要:目的:初步观察肝素在高糖影响大鼠腹间皮细胞(Rat peritoneal mesothelial cells,RPMC)增殖及其表达白细胞介素-1,(Interleukin-l,IL-l)中的作用,从而为持续性非卧床腹膜透析(Continuous ambulatory peritoneal dialysis,CAPD)中肝素的临床应用打下一定的理论基础。方法:建立RPMC的体外培养;并用四四氮唑蓝(MTT)细胞增殖实验和酶联免疫吸附方法(ELISA),来分析肝素对高糖(3.86%葡萄糖)影响RPMC增殖及其分泌IL-l的作用。结果:3.86%葡萄糖能显著抑制RPMC的增殖,而1.25U/ml肝素能部分逆转高糖对RPMC增殖的抑制作用(P <0.01),3.86%葡萄糖能显著增加RPMC表达IL-1β,而1.25U/ml肝素能部分减少高糖增加RPMC表达IL-1β的作用(P<0.01)。结论:本文结果提示,肝素可能通过逆转高糖对RPMC的增殖抑制作用及减少高糖增加RPMC表达促炎细胞因子IL-l的作用,而延缓CAPD相关性腹膜纤维化的发生,最终有益于CAPD进行。Objective To investigate the effect of heparin on peritoneal mesothelial cell proliferation (PMC) as well as its expression of interleukin-1 (IL-1) influenced by high glucose in rats in order to provide some theory basis for the clinial use of heparin during continuous ambulatory peritoneal dialysis (CAPD).Methods PMC were cultured in vitro and MTT method was used to observe the effects of both high glucose (3.86%) and heparin (1.25U/ml) on cell proliferation.ELISA analysis was performed to observe the change in IL-1 of the culure media. Results In the rats high glucose of 36.86% had significant inhibitory effect on PMC proliferation (P <0.01),which was partly reversed by heparin (1.25U/ml), and cpuld induce the expression of LL-1β (P <0.01),which was partly decrease by the heparin.conclusion Heparin can retard the progress of peritoneal fibrinosis related with CAPD and is beneficial to the process of CAPD probably by reversing the inhibitory effect of high hlucose on PMC proliferation and decreasing the pro-motive effect of high glucose on PMC expression of the IL-1
