摘  要:目的将胎兔雪旺细胞种植到脱细胞的同种异体神经段中制成神经桥接复合体,修复神经缺损。方法实验包括胎兔雪旺细胞培养、去细胞神经桥接体的制备以及去细胞神经桥接体复合体的动物实验3个部分。使用双酶消化法培养雪旺细胞并将其种植于经3%TritonX-100作用96 h后的同种异体神经中。实验组,将胎兔雪旺细胞神经复合体桥接移植到兔缺损的坐骨神经上。对照组,移植未注入雪旺细胞的去细胞同种异体神经段。术后观察坐骨神经的再生及功能恢复情况,观察兔足部溃疡形成及愈合情况。于4、8、12周行肌电图检查,在光镜下观察神经轴突的再生情况,并进行统计学分析。结果术后4、8、12周2组动物手术区局部均未出现明显的排斥反应,实验组足部溃疡愈合情况、再生神经纤维数目优于对照组。神经传导速度于术后4周时,2组均未见明显的神经传导,术后8周实验组(24.88±1.19)m/s,对照组(18.64±0.75)m/s(t=-10.92,P<0.05);术后12周实验组(38.66±2.02)m/s,对照组(33.49±1.51)m/s(t=-5.03,P<0.05)。2组差异有统计学意义。神经纤维数目,术后4周实验组(751.67±74.14)根,对照组(541.67±62.42)根(t=-5.31,P<0.05);术后8周实验组(2067.83±233.65)根,对照组(1805.50±155.65)根(t=-2.29,P<0.05);术后12周实验组(3 430.33±128.48)根,对照组(2 491.17±158.95)根(t=-11.26,P<0.05)。2组差异有统计学意义。结论胎兔雪旺细胞神经复合体不但能够为神经再生提供良好的支架作用,而且能够诱导和促进神经轴突的再生。Objective To plant fetus rabbit Schwann cells(Scs)into the extracted acellular nerve of homogeneity and make into a nerve bridging-complex.To repair defective sciatic nerve with the complex.Methods This experiment includes the following steps:preparation of the Scs, preparation of extracted nerve grafts(eNG)and animal experiment.Enzyme digestion(trypsin/ collagenase)was applied to culture Scs.The Scs were micro-injected into the eNG(treated by 3% Triton X-100 totally for 96 h).Experimental group was transplanted nerve to repair the rabbit defective sciatic nerve.The control group was transplanted extracted nerve graft of homogeneity (without the fetus rabbit Schwann cells).To observe the ulcers on the feet of the rabbit,detect the regeneration of axons of rabbit sciatic nerves with light microscope,and measure nerve conduction velocity(NCV)by electromyogram on 4,8 and 12 weeks after operation.Results On the fourth,eighth and twelfth week after operation,the rejection was not found in the operating field of two groups' animal.The experimental group was better than the control group in healing of ulcers and in the number of nerve fibers.On the fourth week after operation,the NCV were not different between the two groups.Eighth week after operation,the NCV were(24.88±1.19)in the experimental group,(18.64±0.75)in the control group(t=-10.92,P<0.05);and twelfth week after operation NCV was(38.66±2.02)in the experimental group and(33.49±1.51)in the control group(t=-5.03,P<0.05).There was significant difference between the two groups.The number of nerve fibers,Fourth week after operation,the number of nerve fibers were (751.67±74.14)bundles in experimental group,(541.67±62.42)bundles in the control group( t=-5.31,P<0.05).Eighth week after operation,(2 067.83±233.65)bundles in experimental group,(1805.50±155.65)bundles in the control group(t=-2.29,P<0.05).Twelfth week after operation,(3 430.33±128.48)bundles in experimental group,(2 491.17±158.95)bundles in the control group(t =-11.26,P<0.05).There was significa
