摘 要:目的 :探讨败血症大鼠不同时间血浆促炎性细胞因子 (IL 8、TNF α)及外周血液和腹腔渗出液中炎性细胞的变化规律。方法 :采用盲肠结扎加穿刺法复制大鼠败血症模型 ,分别在 0、8、1 6h取血和腹腔渗出液 ,用酶联免疫吸附试验 (ELISA法 )测定血浆IL 8、TNF α含量 ;用光镜计数外周血和腹腔渗出液中的白细胞数。结果 :败血症组大鼠 8、1 6h血浆IL 8、TNF α水平均较 0h明显增高 (P <0 0 1 ) ,且以 8h增高更为显著 ,败血症组大鼠血浆IL 8、TNF α在 8、1 6h均明显高于假手术对照组 8h和 1 6h水平 (P <0 0 5 ) ;败血症组大鼠 8、1 6h外周血白细胞计数均较 0h明显降低 (P <0 0 1 ) ,且呈递减趋势 ,败血症组大鼠 8、1 6h外周血白细胞计数均明显低于假手术对照组 ;败血症大鼠腹腔渗出液中白细胞计数 8、1 6h较 0h明显增高 (P<0 0 1 ) ,且呈递增趋势 ;败血症组大鼠 8、1 6h腹腔渗出液中白细胞计数均明显高于假手术对照组。结论 :IL 8、TNF α在大鼠败血症发生发展过程中可能起重要作用 ,败血症大鼠外周血和腹腔渗出液中炎性细胞异常分布与IL 8、TNFObjective:To detect the changes of facilitative inflammatory cell factors (IL 8,TNF α) in blood plasma and inflammatory cells in peripheral blood and abdominal effusion at different periods of time in septicemic rats.Methods:Septicemia models were replicated by means of cecum ligature puncture,the levels of IL 8 and TNF α were measured by enzyme linked immunosorbent assay (ELISA) and the leucocytes in peripheral blood and abdominal effusion were counted under microscope after rats' blood and abdominal effusion were taken at 0,8 and 16 hours post experimentally.Results:In septicemia group,levels of IL 8 and TNF α at 8 and 16 hours after the experiment were noticeably higher than those at 0 hour ( P <0 01) post experimentally with that at 8 h more evident;IL 8 and TNF α levels of septicemia group at 8 and 16 hours were obviously higher than those of control group on which sham operations were done ( P <0 05).The number of leucocyte in peripheral blood of septicemic rats at 8 and 16 hours post experimentally was much lower than that at 0 hour ( P <0 01) with a trend of progressive decrease ,and the number in abdominal effusions had a trend of progressive increase ( P <0 01).Conclusion:IL 8 and TNF α may play a major role in the course of rats' septicemia and the abnormal distribution of inflammatory cells of septicemic rats is closely related to IL 8 and TNF α.
