高效液相色谱法测定人血浆中奥美拉唑对映体的浓度

摘　　要：考察了奥美拉唑对映体在多糖类手性色谱柱(Chiralpak AD柱、Chiralcel OJ柱、Chiralpak AS柱和Chiralcel OD柱)上的分离行为及流动相组成对对映体分离的影响,建立了血浆中奥美拉唑对映体的分析方法。血浆样品采用Agilent BON DODS-C18固相萃取柱提取,以Chiralpak AD(250mm×4.6mm,10μm)为色谱柱;流动相:V(正己烷):V(乙醇)=30:70;流速:0.5mL/min;检测波长:302nm;柱温:25℃。方法的相对回收率为(-)-(S)-奥美拉唑96.7%～100.4%,(+)-(R)-奥美拉唑97.4%～100.2%;日内RSD<4.7%,日间RSD<3.3%;奥美拉唑对映体在5～1000ng/mL范围内线性关系良好(n=5,r=0.9997),最低检测浓度(S/N=3)2ng/mL。方法可用于奥美拉唑对映体人体药代动力学研究。The enantioselective separation of omeprazole enantiomers on different polysaccharide-based chiral stationary phases (Chiralpak AD column,Chiralcel OJ column,Chiralpak AS column and Chiralcel OD column) were evaluated and the effects of mobile phases on enantiomer separation were also investigated.In this assay,a highly sensitive and selective HPLC method for the analysis and quantification of omeprazole enantiomers in human plasma was established.Agilent BOND ODS-C18 solid phase extraction (SPE) cartridges...
