地黄饮子含药脑脊液对Aβ25-35诱导的PC12细胞损伤后生存状态和活力的保护作用

摘　　要：目的:探讨古方地黄饮子对β淀粉样蛋白(Aβ25-35)所致PC12细胞损伤后生存状态和活力的保护作用。方法:把离体培养的进入对数生长期的PC12细胞分为6组:空白对照组、模型对照组、VitE脑脊液组、地黄饮子脑脊液低、中、高剂量组。待24h细胞贴壁后吸去培养液,分别加入空白培养液、正常脑脊液、VitE脑脊液、中药脑脊液低、中、高剂量,加培养液补至等量,37℃孵育2h。然后除空白组之外的各组加入经老化处理的Aβ25-35(终浓度为10μmol/L),建立AD细胞模型。空白组加入等量的培养液,继续孵育24h后收集培养液,对培养液进行MTT法和LDH法检测细胞生存活力。结果:地黄饮子能升高MTT代谢率,降低LDH释放。结论:地黄饮子能改善Aβ25-35损伤后的PC12细胞生存状态,提高细胞生存活力。Objective:To explore the effect of the defined formula of Di HuangYinZi(DHYZ)in regulating and controlling on the survived condition and the vitality in cell PC12 induced by β-amyloid25～35 protein(Aβ25～35).Methods:Cultured Cells pc-12 were divided into 6 groups:normal group,model group,vitamin E group,low dose of TCM group,moderate dose of TCM group and high dose of TCM group.Six groups were respectively added to normal cultured fluid;nomal cerebrospinal fluid,vitE cerebrospinal fluid,low dose of TCM cerebr...
