摘 要:目的 研究大鼠SCI后VEGF在脊髓神经细胞的表达情况。方法 SD大鼠 32只随机分为椎板切除组和脊髓损伤组 ,采用Allen法从背侧致伤T10脊髓 ,于术后 8h ,1,2 ,3d ,1,2 ,3周切取脊髓 ,S -P法免疫组化染色 ,观察VECGF的分布 ,对神经细胞的阳性表达率进行定量分析。结果 对照组脊髓中 ,无VEGP的表达 ,SCI后 8h出现VEGF强表达 ,1~ 3d达到高峰 ,并持续 1周。 2 ,3周后 ,VEGF表达率大幅度下降 (P<0 .0 1) ,前角大运动神经元较其他神经细胞阳性表达率明显增高 (P <0 .0 1) ,背侧白质中胶质细胞较灰质和腹侧白质胶质细胞阳性表达率高 (P <0 .0 1)。结论 大鼠SCI早期 。Objective: To study the expression of vascular endothelial growth factor (VEGF) in spinal nerve cells after SCI in SD rats. Methods: 32 SD rats were divided at random into laminectomy and injury groups. The rats in injury group were injured in dorsal side of T 10 spinal cord by Allen's stall. The rats were sacrificed at 8 h, 1 d, 2 d, 3 d,1 w, 2 w and 3 w after surgery to sample spinal cord. Immunohistochemistry (SP) was performed to observe the distribution of VEGF in the spinal cord.Results: No VEGF immunoreactivity existed in normal spinal cords. Strong VEGF immunoreactivity was observed at 8 h after SCI. It peaked from 1 and 3 days and continued for 1 week.After 2 to 3 weeks immunoreactivity decreased significantly ( P <0.01).VEGF positive rate of large motor neurons was higher than that of other kinds of nerve cells ( P <0.01). The positive rate of glial cells in dorsal white matter was higher than those in gray matter and ventral white matter ( P <0.01). Conclusion: The expression of VEGF in nerve cells in the injured spinal area was seen in the early stage of SCI in rats.
