摘 要:目的探讨经海马组织免疫过的大鼠大脑的病理改变,为研究海马功能提供一种新的实验方法。方法以新鲜豚鼠海马组织匀浆为抗原免疫SD大鼠建立动物模型,进行行为学观察以及用免疫组织化学染色方法检测即刻早期基因Arc/Arg3.1的表达情况。结果行为学观测到模型组大鼠存在异常兴奋,易激惹的表现,而对照组未见明显异常;免疫模型组海马CA3区有明显的Arc/Arg3.1阳性细胞表达,高于正常对照组P<0.05;而在对照组和模型组的海马齿状回均未见明显的Arc/Arg3.1阳性细胞。结论我们推测模型组大鼠的异常兴奋和易激惹表现可能与海马匀浆中的某些抗原刺激有关,而Arc/Arg3.1表达的增加可能是这种刺激在海马产生一系列生物化学反应的标志。Objective To investigate the pathological changes in rat brain after immuned by hippocampal tissue,and provide a new experimental method for the study of hippocampal function.Methods The fresh guinea pig hippocampal tissue was applied as antigen to immune SD rats for establishment of animal models,behavioral observations and the use of immunohistochemical staining were done to detect the expression of Arc/Arg3.1.Results Behavior observed abnormal excitement,irritability performance in model group,while no obvious abnormalities in control group.In hippocampal CA3 area of the model group,the expression of Arc/Arg3.1 positive cells was higher than that in the normal control group.The hippocampus dentate gyrus had no obvious expression of Arc/Arg3.1 positive cells in both control and model groups.Conclusion The abnormal excitement and irritability of the rats in model group may be related to the performance of some of the hippocampal homogenate antigen stimulation.The increased expression of Arc/Arg3.1 may be a marker for a series of biochemical reactions by the stimulation in the hippocampus.
