小檗碱对宫颈癌Hela细胞Caspase-3基因表达的影响

摘　　要：目的:研究小檗碱(berberine)对人宫颈癌Hela细胞体外增殖、凋亡及凋亡相关基因Caspase-3表达的影响。方法:MTT法测定不同浓度(0～40μmol/L)berberine干预Hela细胞后的凋亡率;RT-PCR检测Caspase-3mR-NA表达水平。结果:berberine呈剂量-时间依赖方式抑制Hela细胞的生长(P<0.01);且随着berberine浓度增高,细胞凋亡增加,Caspase-3mRNA表达上调(P<0.001)。结论:berberine抑制人宫颈癌Hela细胞生长、诱导凋亡的机制可能与上调Caspase-3mRNA表达有关。Objective To investigate the effects of berberine on inducing the proliferation,apoptosis and expression of Caspase-3 gene in human cervical cancer Hela cell line in vitro.Methods The apoptosis rate of Hela cells were determined with MTT method after treating with different concentrations of berberine(0~40 μmol/L).The expression of Caspase-3 mRNA were detected by RT-PCR.Results The inhibitory effect of berberine on the growth of Hela cell lines was in a time and dose dependent manner.The apoptosis rate and Caspase-3 mRNA expression were obviously increased following the increasing concentration of berberine(P﹤0.01).Conclusion Berberine could inhibit proliferation and induce apoptosis of Hela cells,which may be related to the upregulation of Caspase-3 mRNA expression.
