益肾调督针法对脑缺血再灌注大鼠P-selectin mRNA和蛋白表达的影响

摘　　要：目的:通过观察益肾调督针法对脑缺血再灌注大鼠脑内P-selectin mRNA和蛋白表达的影响,探讨益肾调督针法治疗缺血性脑损伤的机制。方法:采用大鼠局灶性脑缺血再灌注模型,应用原位杂交及免疫组化方法观察脑缺血再灌注时P-selectin mRNA和蛋白表达的变化,以及针刺对其影响。结果:正常组和假手术组大鼠P-selectin mRNA和蛋白在皮质区未见表达,脑缺血再灌注后12 h P-selectin mRNA和蛋白表达增强(P<0.05),针刺可明显抑制脑缺血区皮质P-selectin mRNA和蛋白的表达(P<0.05或0.01)。结论:益肾调督针法对缺血性脑损伤的保护作用机制与针刺抑制脑内P-selectin mRNA的表达从而减少P-selectin蛋白的合成有关。Objective:This study was designed to observe the effect of Yishen Tiaodu acupuncture manipulation on the expression of p-selectin mRNA and protein in cerebral ischemia-reperfusion rats and to further investigate the protective mechanism of Yishen Tiaodu acupuncture manipulation against ischemic brain damage from the angle of molecule in order to reveal deeply the mechanism of acupuncture treatment on cerebral ischemia.Methods:With the randomized controlled design and a random number table,Wistar rats were divided into normal group,sham operation group,model group,Yishen Tiaodu acupuncture group and common acupuncture group(30 cases for each group).MCAO model was produced by left middle cerebral artery occlusion by suture method in the rats.Experimental rats in each group were given corresponding intervention and were sacrificed at 12,24,and 48 hours respectively.After specimen handling,immunohistochemistry and hybridization in situ were used to check the expression of p-selectin mRNA and protein in rat brain tissue.Results:Experimental results showed the expression of p-selectin mRNA and protein in rat brain tissue were increased significantly after focal cerebral ischemia at 1 hours and reperfusion at 12 hours in model group.The expression of p-selectin protein were at the peak at 24 hours and then were decreased gradually and still higher significantly then the normal group at 48 hours.The expression of p-selectin mRNA were comparatively high and then showed decrement gradually in 12 hours.Compared with the sham operation group and normal group,the expression of p-selectin mRNA and protein in rat brain tissue in the model group were significantly different(P<0.01) in each time.Compared with other groups,after acupuncture treatment the expression of p-selectin mRNA and protein were decreased significantly(P<0.05,P<0.01) in the Yishen Tiaodu acupuncture group in each time.It meaned the expression of p-selectin mRNA and protein in rat brain tissue were raised significantly after focal cerebral ischemia but they we
