摘  要:目的:观察不同时间点电针对大鼠脑梗死后神经干细胞增殖、分化的影响,探讨其神经康复机制和最佳时机。方法:采用线栓法制备大鼠MCA0模型,随机分为对照组和电针治疗组(各组分1、3、7、14、21、28天时间点,n=10),原位杂交法半定量分析FGF-2 mRNA表达情况,通过横木实验测试运动整合变化。结果:MCAO后1、3、7、14、21、28天6个时间点,电刺激治疗的大鼠运动功能的恢复较对照组明显改善(P<0.05),但在治疗组内21、28天时间点比较无明显差异(P>0.05),FGF-2 mRNA表达水平电针治疗组与对照组相比在各个时间点早期存在显著性差异(P<0.05)。结论:电针可以促进脑梗死后内源性FGF-2 mRNA合成增加,早期及时对神经功能康复尤为重要。Objective:To observe the effects of electroacupuncture on the express levels of FGF-2 mRNA,and to explore the possible mechanism and optimal treatment time of electroacupunctue in treatment of ischemic stroke.Method:Animal models were made by ligating external carotid artery and inserting a piece of nylon thread into the internal carotid artery among 120 Wistar rats.Then the rats were randomly devided into treatment group and control group.(each group contains 1、3、7、14、21、28d six time points,each point n=10);After seven days the two group MCAO rats of the motor function were evaluated by using the beam walking test(BWT),then all animals were sacrificed after different durations,FGF-2 mRNA levels were determined by in situ hybridization,the results were analyzed by micro-image analysis system.Result:At different time points after cerebral ischemia,motor function was significantly improved in the treatment groups with electric stimulation compared with the control group(P0.05),the express levels of FGF-2 mRNA were significant between the treatment groups and the control groups at early time points(1、3、7、14d)(P<0.05).Conclusion:Electroacupuncture can increase express levels of FGF-2 mRNA after cerebral ischemia.It may be one of mechanisms of electro acupuncture in treating ischemic stroke.
